Tim,
If you see no diagonal lines - then remove NOTHING.
Yes there is a film on both sides of the diffuser, and that is a benefit.
If the rear side has the diagonal lines, then that film should be removed
because the stripes show through.
There is no protective film on the back side of the LCD - it is only on the
front side. You should remove the front film just before putting the front
panel in place - until then, it will protect the LCD surface from scratches
wile you are working on the assembly.
